We had a can of prunning paint and I thought that's what I grabbed. It turned out to be craft enamel. If I leave it on will it kill or damage the tree?

I am unsure what the chemical makeup of craft enamal is but I will say this: if it is harmful, the damage has been done. If it is a large tree I highly doubt you would cause it stress even if it turned out to be harmful. Dried paint shouldnt bother a tree in the least. Just leave unless it looks ugly. like hot pink or something. Then spray a more natural color over it. At least thats what I would do.
