Hi, my name is Christine and I'm from Missouri. I am currently taking a
horticultural class at our local college. For this class we are to study,
identify and have a leaf sample for 130 different trees. Yes, the Giant
Sequoia is on that list and yes, I am having a very difficult time, to say
the least in locating a "piece" of this tree! I am wondering if you can
help me by sending a portion (stem and needles) of this tree to me or
direct me to someone that may be able to help me in my search? I
appreciate your time.

If there is an Arboretum in your area , they may have a Sequoia, but they are usaully not happy to let you cut a twig off . We have several specimens near here at Longwood Gardens and Tyler Arboretum. John
 
Boerner Gardens here in MKE County had someone neatly cut 2 inch squares out of the bark of a number of specimine trees. Each cut at around 4ft. They concidered going to all the local schools to find if any kids did a project. It was a few years back.

It is against the law to coolect samples form any of the plants without a permit. The fines are pretty steep.
